For designing and preparing synthetic cells from the bottom-up, a circular workflow from computer-aided system design and assembly of cell constructs to experimental validation and systematic optimization should be established. In doing so, one should establish libraries of well-characterized functional biomolecular units and pursue a modular approach on different system hierarchies.
